Create Designer Exosomes with XPack Lentivectors
SBI XPack技术可以实现将兴趣蛋白包装进exosomes中,获得的exosome可直接用于处理靶细胞。SBI优化了一个特殊的多肽序列,可将蛋白介导至exosome内膜上,此融合蛋白即被包装进exosome中,并随exosome分泌至细胞外。这一“XPack”多肽序列已包含在XPack慢病毒表达载体系列中,以保证报告基因蛋白或者其他兴趣蛋白被包装入exosome中,并随之分泌细胞外。这些经改造的exosomes可以通过分离富集,随后运输到靶细胞中。
选择合适的XPack reporter lentivectors或者pre-loaded reporter exosomes,可实现通过荧光或者发光反应来动态观察exosomes。此外,研究者改造含有兴趣蛋白的exosome,并研究exosome的靶细胞运输。
